What is the method of preperation to use this product?
This product is provided as a lyophilized powder. You have to reconstitute it in 250 µl of DMSO. This makes it 250 µl of 500X inhibitor cocktail. Which means that for every 499 or 500 µl of the lysate you can add 1 µl of the inhibitor. This means that the total amount of this inhibitor i.e., 250 µl is enough for 500 x 250 µl of the lysate, which is 125 ml of lysate. Therefore the dilution factor is 500.

Will DMSO is provided with the cocktail? While preparing 1X from 500X, what is used as a diluent (DMSO or water)
No, DMSO is not included with this cocktail. You need to procure anhydrous molecular biology gradient DMSO for this. The diluent used for the 500X to 1X dilution can be either the media, or the lysis solution/buffer or even PBS. It should not be DMSO.
