Antibody and protein aggregation are important issues in the field of Biotherapeutics. A wide range of conditions and different stages of processing of biopharmaceuticals, such as, formulation, purification, storage, shipping and handling, heat, freeze thaw cycles, mechanical stress, chemical treatment etc. might cause aggregation of proteins, enzymes and antibody targets. The possibility of protein aggregates in eliciting immunogenicity is a major concern for biotherapeutics, which might compromise the safety and efficacy of the biologically derived drug products. Recombinant protein production and storage would highly benefit from a high throughput screening method that can screen a variety of formulation and storage conditions of a particular target protein/antibody. Currently, different biophysical techniques such as Dynamic Light Scattering (DLS), Size-exclusion Chromatography with Multi-angle Light Scattering (SEC-MALS) can detect the presence of small amount of aggregates in a protein/antibody preparation, but they are not amenable for high-throughput screening format. Can I use this kit on protein lysates from tissues and cell lines?
You can only use this kit on purified antibody, protein or enzymes and not on crude extracts.
Can the kit work on bacteria or yeast cells?
The kit has been standardized for mammalian cells only
What is the exact volume of sample required for this assay?
There is no specific volume we can recommend for the amount any sample to be used since it is completely sample concentration and quality based. You have to do a pilot expt with multiple sample volumes to determine the optimal volume which gives a reading within the linear range of the standard curve. Can we use an alternate buffer for sample preparation (cell lysis, sample dilutions etc)?
Our assay buffers are optimized for the reactions they are designed for. They not only contain some detergents for efficient lysis of your cells/tissue, but also contain some proprietary components required for the further reactions. Therefore, we highly recommend using the buffers provided in the kit for the best results.
