Description




Lysosomes are membrane-bound cell organelles found in most animal cells (except red blood cells). Lysosomes are spherical vesicles containing hydrolytic enzymes capable of breaking down different kinds of biomolecules, including proteins, nucleic acids, carbohydrates, lipids, and cellular debris. They are known to contain over 50 different enzymes, which are active under acidic conditions. Thus, lysosomes act as the waste disposal system of the cells by digesting unwanted materials in the cytoplasm. BioVision’s Lysosome Isolation Kit provides a procedure for isolating enriched or purified lysosomal fraction from animal tissues and cultured cells by differential centrifugation followed by density gradient centrifugation

Can we use flash frozen tissue for isolating lysosomes?
No. Isolation of lysosomes is recommended from fresh samples and not frozen samples.

I only have 10 ml ultracentrifuge tubes and not 2 ml tubes as suggested in the datasheet. Can I use a 10 ml ultracentrifuge tube instead of a 2 ml volume?
Yes, 10 ml tube can be used. But for preparing the density gradient for a 10 ml tube, the volumes given for 2 ml gradient must be multiplied by factor 5. Also, we recommend increasing the starting cell number from 2 X10^7 to 3x10^7.

What is the exact volume of sample required for this assay?
There is no specific volume we can recommend for the amount any sample to be used since it is completely sample concentration and quality based. You have to do a pilot expt with multiple sample volumes to determine the optimal volume which gives a reading within the linear range of the standard curve. Please refer to the citations for this product to see what other clients have used with similar sample types.

Can we use an alternate buffer for sample preparation (cell lysis, sample dilutions etc)?
Our assay buffers are optimized for the reactions they are designed for. They not only contain some detergents for efficient lysis of your cells/tissue, but also contain some proprietary components required for the further reactions. Therefore, we highly recommend using the buffers provided in the kit for the best results.
